An Evolutionary Intermediate of DNA Methyltransferase for the Epigenetic Control

Abstract
DNA methyltransferase family is responsible for the epigenetic control of genome, especially critical for the incidence of cancer. We have developed combinatorial protein fragment library of DNA methyltransferase M.HhaI and screened for the functional pair of splitted DNA methyltransferase in E.coli. Using this procedure, several splitted methyltransferases were identified. Exploring the evolution of naturally permutated DNA methyltransferase family, this process can be used to identify active evolutionary intermediates of DNA methyltransferase which might be useful for cancer therapy and cancer prevention.
